Sixteen of the letters are from the Orthodox Greek theologian Metrophanes Kritopoulos (1589-1639), who was later to become Patriarch of Alexandria. The letters contained in the manuscript date from this period and are addressed to English scholars, such as Andrew Downes (ca. 1549-1628), classical scholar, fellow of St John's College, Cambridge, and Patrick Young (1584-1652), Scottish chaplain of All Souls’ College in Oxford and later royal librarian.

Along with Kritopoulos' letters, the manuscript contains ten letters from the French classical scholar Isaac Casaubon (1559-1614).
